I had exactly this problem... it is because DNN stores the alias for a portal. So if you first run the application under VS then it stores the alias as localhost:1234/nuke (where 1234 is the port number). Then when you try and run it under IIS at locahost/nuke it internally tries to redirect to localhost:1234/nuke unless you have added locahost/nuke as another alias.

If you can still get into DNN as a host then login and add the extra alias. If you can't then you need to open the database and edit table PortalAlias and change it the the correct URL.
