Host and Admin Tabs Malfunction

This is kinda weird,
I have adev environment setup on my local computer accessing a SQL DB on another server. 
Initially all this works. I am able add pages etc...
Yet each time I add the telerik library and test running Linq, certain links just stop working forexample on module definitions selecting create module definition forwards me back to the list. Trying to edit any of the definitions does the same.

Am really quite stumped as there are no errors registered for this it just stops working but seems to think that its doing a fine job.

EDIT: Okay so now no admin navigation works also adding an alias to the portal doesn't take affect.
I am running Vista on my local machine but I can't see why this should make any difference considering I've see others running IIS 7 without issue.

Any ideas?

I have tracked my issues down to the following assemblies and I'm now going to start to add them one at a time to see which cuase the issues

Okay have narrowed it down as soon as I add

 <add assembly="System.Web.Extensions, Version=3.5.0.0, Culture=neutral, PublicKeyToken=31BF3856AD364E35" />

It all goes potty... I'm still farily new to .NET only a year nnow but why is that the case?

you'll need to download and install the 3.5 version of .net (see http://www.chrishammond.com/tabid/54/itemid/970/DNN-4X-development-in-Windows-Vista-issues-with.aspx for link)

Afriad thats not solved the problem see the 1.0.61025.0 version of web.extensions obviously can't work along side the 3.5 version but if the 3.5 version is installed dotnetnuke keels overs as far as I can see this is not a vista specific issue I've now tried it on Server 2003 too. to get DNN to work with the 3.5 version should I recompile to source with the 3.5 reference?
I'm assuming that it is the Client API that is relying on this assembly

you don't need to recompile. If you've installed 3.5 ajax (and it's extensions) will be in the GAC, so you can delete it from your bin folder. However, your web.config will have to contain the correct targetted version i.e You'll need to change the version numbers to Version=3.5.0.0 (From Version=1.0.61025.0) if you're targetting version 3.5 of the framework.
