Mike,  my issue is not the same as the others.  I had 3 out of like 15 security roles stop working abrubptly using version 1.0.0 0. when it had been working for over a year.   I have 5 regions with 3 roles a piece and one region basically stopped syncing roles but login was fine.

Yeah I know it was different but I've got a feeling (based on other posts) that it's going to end up being tied to how I was doing the nested group search. I've had other users who had it working perfectly under .03 suddenly have the roles no longer syncing. I haven't had a chance of going back and checking but I suspect there may have been an update put out by Microsoft that hosed my code.

I can't remember if I've mentioned this before but have you tried the LDAPBrowser (http://www.ldapbrowser.com/download)? I've got it installed on my dev system and use it to connect to the AD using the same credentials I'm using in DNN to see what information I should be able to pull in. It's my "back-up" when I don't think my code is returning correct/complete information.

Mike I am was still using 1.0.0.0 on my production where this is happening.

Doh!!! I didn't realize you were still using 1.0.0. So my nested group search theory doesn't apply. In that case I'm at a loss. Why it would be working and suddenly stop has got me stumped. Is there a chance of setting up a mirror site that you can mess around with without the worry of it blowing up?
