I'm trying to setup an AD on a DNN 5.4.1 with AD 5.0.2 and I get this error on the config page

(system is in spanish: Error desconocido => Unknown Error)

System is a Windows Server 2008  SP2.

Usually this happens when the site cannot access the Active Directory. A couple of things to check; is the server on the same domain as the AD?, Are you running the site in the Classic pipeline?, Have you tried using impersonation in the web.config?

Mike, the site is running on asp.net 2 Integrated. I've asked if it's on the same domain as the AD, I think it's but I'm not sure.

And no, I've not yet tried to use impersonation, do you think this is what I should try?

BTW, I clicked UpdateSettings and I saw the message sayin the domain was found.
But I still get this error message on the screen.

At some point this was working on a Win2003, now the site moved to a Win2008 SP2 64 and I can still login with users already created but no new user is able to login to the site.

The 5.0.2 version of the provider won't work properly under the Integrated Pipeline so I'm pretty sure that's the cause of your problems. See this post by Dan Ball on how to get it working on Server 2008. http://www.dotnetnuke.com/Resources/Forums/forumid/89/threadid/402659/scope/posts.aspx
